Replication of all positive-sense single-stranded RNA viruses occurs in specific structures in close association with cellular membranes. Targeting of the viral replication complex (RC) to the site of replication is mediated by the interaction of viral-encoded proteins and host factors. Electron microscope studies have shown that Poinsettia mosaic virus (PnMV, family Tymoviridae) infection is associated with the presence of vesicular structures in the chloroplasts, which indicates that the replication of PnMV might occur in association with chloroplast-derived membranes. Using computer assisted homology search, we have identified that the coat protein (CP) of PnMV shows similarity to membrane bound proteins and contains a conserved amino acid sequence motif found in members of the Alb3/Oxa1/YidC protein family. This protein family is involved in the insertion of proteins into intracellular membranes. In this study we carried out localization studies combined with confocal laser microscopy to identify the cellular localization of the PnMV CP. Transient expression of red fluorescent protein (RFP)-tagged PnMV CP in Nicotiana benthamiana protoplast was shown to localize in the chloroplast.

Epigenetic memory formed in the Norway spruce embryos permanently affect the timing of bud burst and bud set in the progenies, vitally important adaptive traits in this long-lived forest species. Epigenetic memory marks are established in response to the temperature conditions prevailing during embryogenesis; the epitype is fixed by the time the embryo is fully developed and is mitotically propagated throughout the tree’s life span. Somatic embryogenesis closely mimics the natural zygotic embryo formation and results in epigenetically different plants in a predictable temperature-dependent manner with respect to altered phenology. Using RNAseq transcriptome analysis of mRNA and noncodingRNA (ncRNA) changes were monitored in somatic embryos under different temperatures. We found distinct differences in mRNA and ncRNA transcriptomes between the genetically identical embryogenic tissues grown under the epitype-inducing temperatures suggesting temperature-dependent canalizing of gene expression during embryo formation, putatively based on chromatin modifications.

Ten saplings of European ash (Fraxinus excelsior L.) naturally infected by the invasive ash dieback pathogen Hymenoscyphus fraxineus were collected in Ukraine and Norway and examined for bark necrosis and extension of discoloration in sapwood and pith in a stem region. Tissue-specific colonization profiles were determined by spatial analyses of symptomatic and visually healthy stem tissues using a H. fraxineus-specific qPCR assay and light microscopy. Our data suggest that hyphal growth in the starch-rich perimedullary pith is of particular importance for both axial and radial spread of H. fraxineus, but that most of its biomass accumulates in sapwood parenchyma. The study confirms the results from earlier work and presents new information that refines the current stem invasion model.
