European canker is one of the most devastating fungal diseases of apple in most temperate regions. The causal agent, Neonectria ditissima, infects trees through wounds in the bark forming cankers that girdle the stem and eventually cause tree death. Timely protection of the trees is challenged by stagnation of symptom expression after infections for a long period of time. The objective of this research is to use a novel TaqMan PCR assay to detect and quantify N. ditissima during the asymptomatic colonization of apple wood. Pruning wounds on branches of the cultivars Elstar and Gala were inoculated with N. ditissima and wood discs were sampled at 2–6, 10–14, and 30–34 mm distance from the inoculation site after 3 hours, 2 weeks, 4 weeks and 8 weeks for the detection and quantification of the pathogen. The TaqMan PCR assay detected N. ditissima in 51% of the inoculated apple tree samples. This was more sensitive than the culturing method detecting N. ditissima in 11% of the samples. An accumulation of N. ditissima DNA up to 34 mm distance from the inoculation site was observed without development of visible symptoms. To our knowledge this is the first time colonization of N. ditissima was detected and quantified in the absence of symptoms of European canker. The implications of this research are discussed.

The effect of steam thermotherapy on Botrytis spp. populations in strawberry transplants was evaluated. Tray plants rooted in 0.2 L peat plugs of seasonal flowering cvs. Falco, Sonsation, and Soprano, and everbearing cvs. Favori and Murano were pre-treated with steam at 37 °C for 1 h, followed by 1 h at ambient temperature and air humidity, and then 2 or 4 h steam treatment at 44 °C. Except for one cultivar with a slight reduction in yield, there were no negative effects on plant performance. Compared to untreated transplants, mean incidence of Botrytis on the five cultivars was reduced by 43 and 86% with the 2 and 4 h treatments, respectively. Within cultivars the reduction was significant in 2 and 3 experiments following the 2 and 4 h treatments, respectively. Sclerotia from four different isolates of Botrytis were subjected to treatment including 4 h of steam thermotherapy and subsequently tested for viability. Following 14 days of incubation, 90 to 100% (mean 97%) of treated sclerotia failed to produce mycelial growth compared with untreated sclerotia, which all germinated and produced mycelia. Botrytis isolates recovered from both treated and untreated strawberry transplants were tested for resistance to seven fungicides, including boscalid, fenhexamid, fludioxonil, fluopyram, pyraclostrobin, pyrimethanil and thiophanate-methyl. Multiple fungicide resistance was common; 35.5% of isolates were resistant to fungicides from at least three FRAC groups. Results indicate that steam thermotherapy treatment strongly reduces populations of Botrytis spp., including fungicide-resistant strains, in strawberry transplants with negligible negative impacts on the transplants.

Overvåkingsprogrammet i 2022 omfattet undersøkelse for tilstedeværelse av furuvednematode (Bursaphelenchus xylophilus) i hogstavfall fra furu og i furubukker av slekten Monochamus. I OK-programmets delaktivitet som omfattet kartlegging av furuvednematode i hogstavfall, ble det tatt 401 flisprøver fra hogstavfall og vindfall av furu (Pinus sylvestris) som hadde tegn på angrep av furubukker i slekten Monochamus. Prøvene ble tatt i Innlandet, Vestfold og Telemark, og Viken. Flisprøvene ble inkubert ved +25°C i to uker før nematoder ble ekstrahert med Baermann-trakt og undersøkt i mikroskop. Furuvednematoden B. xylophilus ble ikke påvist, men den naturlig forekommende arten Bursaphelenchus mucronatus kolymensis ble oppdaget i 16 flisprøver. Siden overvåkingen av furuvednematode startet i 2000, har alle de analyserte flisprøvene, totalt 8924, vært negative for furuvednematode. I OK-programmets delaktivitet som omfattet kartlegging av furuvednematode i furubukker, ble feller med attraktanter for fangst av voksne, flygende furubukker satt opp i Agder, Telemark, Viken (Østfold og Akershus), Hedmark og Trøndelag. Billene ble kuttet i biter og ekstrahert med en modifisert Baermann-trakt. Suspensjonen fra ekstraksjonene ble undersøkt i mikroskop for forekomst av Bursaphelenchus spp. Ingen furuvednematoder ble påvist i de 54 undersøkte billene. Bursaphelenchus mucronatus kolymensis ble ikke heller oppdaget.
