Sammendrag

The challenges of grassland management are not quite the same in different European countries. I have always been of the opinion that there is a lot to learn by visiting other countries and by participating in international conferences. I have on two occasions stayed a year at research institutes in Europe. My first stay was in Switzerland, where I had the opportunity to work on a specific subject; biological nitrogen fixation in white clover. On my second stay, at North Wyke in 1997-98, I had a much broader approach. I wished to work with several of the research groups at the Station in order to learn about research methods and results. All members of the staff had a positive attitude to me as a visiting scientist, and they were helpful and did not seem to be tired of all my questions on research issues, or on social and political conditions in Great Britain.

The application of genetic engineering in the floriculture industry has become instrumental and rewarding in meeting the demand for novel desirable traits. Agrobacterium-mediated transformation for poinsettia (Euphorbia pulcherrima Willd. Ex Klotzsch), a contemporary symbol of Christmas in most parts of the world, is reported here for the first time. Internode stem explants of poinsettia cv. Millenium were transformed by Agrobacterium tumefaciens, strain LBA 4404, harbouring hairpin (hp) RNA gene constructs to induce RNA silencing-mediated resistance to Poinsettia mosaic virus (PnMV). In five experiments utilizing 868 explants, 18 independent transgenic lines were generated. Stable integration of transgenes into the poinsettia nuclear genome was confirmed by PCR and Southern blot analysis. Both single- and multiple-copy transgene integration into the poinsettia genome were found among transformants. Transgenic poinsettia plants showing resistance to mechanical inoculation of PnMV were detected by double antibody sandwich enzyme-linked immunosorbent assays. Northern blot analysis of low molecular weight RNA revealed that transgene-derived small interfering (si) RNA molecules were detected among the poinsettia transformants prior to inoculation. The Agrobacterium-mediated transformation methodology developed in the current study should facilitate improvement of this ornamental plant with enhanced disease resistance, quality improvement and desirable colour alteration. Production of yellow-coloured poinsettia using the Agrobacterium-mediated transformation method is in progress.

Culture of marine finfish species is expanding with emphasis on Atlantic cod production in northern Europe. These new species have their own ectoparasite problems requiring treatment and the use of wrasse is highly successful in controlling sea lice without adding pharmaceuticals to the marine environment (e.g. Deady et al., 1995). To establish a cleaner fish production, the first step is to develop productive broodstock and determine the management practices. In the summer 2008, wild Labrus bergylta were caught to establish an adequate breeding population (Ottesen et al., 2008). These preliminary studies were conducted to identify potential problems regarding the domestication of the ballan wrasse. Fish caught from the wild have difficulty to adapt to transport and / or captivity. As a result, negative indicators have been observed such as interspecies aggression, reduction/lack of maturity, reduced appetite and diseases. The work reported here focuses on this last point and reviews the potential of microalgae to reduce the bacterial disease pressure on L. bergylta in captivity.
