The GH61 represents the most enigmatic Glycoside Hydrolase Family (GH) regarding putative enzymatic activity and importance in cellulose degradation. Heterobasidion irregulare is a necrotizing pathogen and white rot fungus, causing enormous damages in conifer forests.The genome of H. irregulare allowed identification of ten HiGH61 genes. qRT-PCR analysis separate the HiGH61 members into two groups; one that show up regulation on lignocellulosic substrates and another that show either down regulation or constitutive expression. This grouping suggests that the fungus relates different sets of GH61s for different substrates, like in the various stages of necrotizing and saprophytic growth on the host.One HiGH61 showed up to 17000 fold increase on spruce heartwood suggesting a pivotal role in cellulose decomposition during saprophytic growth. Sequence analysis of these genes reveals that all GH61s but one possess the conserved metal binding motif predicted to be essential for activity.The sequences also divide into groups having either an insert near the N-terminus or an insert near the second catalytic histidine, which both may represent extensions of the substrate binding surface. Three HiGH61s encode cellulose-binding modules (CBM1), indicating direct targeting of crystalline cellulose, two being up regulated on pure cellulose.There was a common substrate-specific induction patterns of the HiGH61s with several reference cellulolytic and hemicellulolytic GHs, this taken together with their low levels on media lacking lignocellulose, reflect the concerted nature of cell wall polymer degradation.

Forsøksresultatene som presenteres i denne rapporten er biologisk godkjenningsprøving av ugrasmidler utført på oppdrag fra Mattilsynet i 2012. Inkludert i rapporten er også forsøk eller egne forsøksledd som grupperes som biologisk utviklingsprøving. Forsøkene er utført etter GEP-kvalitet1 hvis ikke annet er nevnt. Dette innebærer at det er utarbeidet skriftlige prosedyrer for nesten alle arbeidsprosesser. Disse prosedyrene, kalt standardforskrifter (SF’er), er samlet i en kvalitetshåndbok. Denne er delt ut til alle personer som arbeider med utprøving av plantevernmidler. De samme personene har også vært med på et endagskurs i GEP-arbeid.

A crucial consideration for strawberry producers in Norway and other northern countries is winter freezing damage. A long-term goal of the Norwegian strawberry breeding is to increase winter hardiness and to improve fruit quality. Due to the complexity involved in regulating and enhancing freezing tolerance, the progress in the improvement of cultivars using traditional screening methods have had limited success. Thus, the development of molecular markers for freezing hardiness would facilitate the selection work for this trait. In this effort, we have developed and adopted state-of-art molecular tools to investigate cold response in strawberry plants during the acclimation phase resulting in the identification of a large number of genes, proteins, and distinct metabolites that correspond to cold/freezing tolerance in strawberry. To identify proteins responsible for freezing tolerance in strawberry we have examined alterations in protein levels in strawberry varieties that differ in cold tolerance using either 2-DE gel analysis followed by LC-MS/MS analysis or a shotgun MS/MS approach. Proteomic analysis suggested 30 potential biomarkers that showed significant changes in the cultivated strawberry in response to cold. In addition, GC-MS-based metabolite profiling revealed the up-regulation of carbohydrates, polyols, amino acids, TCA intermediates, and other distinct secondary metabolites after cold treatment. Transcriptional analysis of the cold acclimated samples also confirmed the regulation upon cold-treatment with varietal differences in strawberry. Moreover, several F2-populations from the model F. vesca parents diverging in cold tolerance have been developed in order to facilitate mapping of QTLs by performing GBS analyses. The knowledge attained from these endeavors is expected to expedite breeding of strawberries to achieve freezing tolerant lines and provide an integrative understanding of the molecular pathways that underlie this characteristic. * Rohloff et al. I 2011 inngikk seks flater i den intensive skogovervåkingen der utviklingen av skogøkosystemet i Norge blir fulgt. Disse flatene inngår i det nasjonale ”Overvåkingsprogram for skogskader”, og de er også en del av det europeiske nettverket av intensive skogovervåkingsflater, der 39 europeiske land deltar med mer enn 800 flater (ICP Forests). Intensiv skogovervåking er utført her i landet siden midten av 1980-tallet, med tidsserier som beskriver endringer i skogøkosystemet. Hovedformålet er å beskrive skogens helsetilstand og belyse virkninger av langtransporterte forurensninger på skogøkosystemet. Observasjonene fra 2011 og utviklingen siden 1986 presenteres i denne rapporten....

Abstract Strict control of morphogenesis is essential in production of potted poinsettia. Commonly, this is obtained by the use of plant growth retardants (PGRs), often in combination with early morning temperature drops. Due to negative effects on human health and the environment, the use of PGRs is becoming restricted. Also, energy-saving growth regimes and periods of high temperatures limit effective use of temperature drops. In the present study the use of a high proportion of blue (B) light provided by light emitting diodes [LEDs, 20% blue (B), 80% red (R)] was compared with traditional high pressure sodium (HPS) lamps (5% B) providing similar phytochrome photostationary state to produce compact poinsettia plants. Both in the greenhouse and growth chamber, all cultivars were 20–34% shorter for LED compared to HPS grown plants. Also, leaf and bract area as well as chlorophyll content and total dry matter accumulation were lower under LED. The LED did not delay bract color formation, visible cyathia and flowering compared to HPS, and no difference in post production performance (cyathia/bract abscission or necrosis) between the two light treatments was found. The effect of end of day-red (EOD-R) lighting combination with LED and HPS supplemental lamps during the photoperiod in the greenhouse was also investigated. Reduced stem extension (13%) was observed under HPS only and for one of the two cultivars tested, whereas under the LED regime, there was no effect of EOD-R lighting.
