Zearalenones are produced by several Fusarium species and can cause reproductive problems in animals. Some aurofusarin mutants of Fusarium pseudograminearum produce elevated levels of zearalenone (ZON), one of the estrogenic mycotoxins comprising the zearalenones. An analysis of transcripts from polyketide synthase genes identified in the Fusarium graminearum database was carried out for these mutants. PKS4 was the only gene with an enoyl reductase domain that had a higher level of transcription in the aurofusarin mutants than in the wild type. An Agrobacterium tumefaciens-mediated transformation protocol was used to replace the central part of the PKS4 gene with a hygB resistance gene through double homologous recombination in an F. graminearum strain producing a high level of ZON. PCR and Southern analysis of transformants were used to identify isolates with single insertional replacements of PKS4. High-performance liquid chromatography analysis showed that the PKS4 replacement mutant did not produce ZON. Thus, PKS4 encodes an enzyme required for the production of ZON in F. graminearum. Barley root infection studies revealed no alteration in the pathogenicity of the PKS4 mutant compared to the pathogenicity of the wild type. The expression of PKS13, which is located in the same cluster as PKS4, decreased dramatically in the mutant, while transcription of PKS4 was unchanged. This differential expression may indicate that ZON or its derivatives do not regulate expression of PKS4 and that the PKS4-encoded protein or its product stimulates expression of PKS13. Furthermore, both the lack of aurofusarin and ZON influenced the expression of other polyketide synthases, demonstrating that one polyketide can influence the expression of others.

When conifers such as Picea abies Karst. (Norway spruce) are attacked by insects or pathogens, they often respond by producing increased quantities of terpenoid oleoresin. This response can be mimicked in young P. abies seedlings by treatment with methyl jasmonate (MJ). In this study, we determined the effects of MJ on terpenoids and other chemical defenses of mature P. abies, and investigated whether this treatment protected trees against attack by the blue-stain fungus Ceratocystis polonica (Siem.) C. Moreau, the most important fungal associate of the spruce bark beetle Ips typo-graphus L. Methyljasmonate treatment induced the formation of traumatic resin ducts in the developing xylem, enhanced resin flow and stimulated increased accumulation of monoterpenes, sesquiterpenes and diterpene resin acids. However, only minor changes were detected in terpene composition in response to MJ treatment and no changes in soluble phenolic concentration were measured. There was much variability in the timing and degree of response to MJ among clones. The observed chemical and anatomical changes in response to MJ treatment were correlated with increased resistance to C. polonica, suggesting that terpenoid oleoresin may function in defense against this pathogen.

In order to improve the basis for utilising nitrogen (N) fixed by white clover (Trifolium repens L.) in northern agriculture, we studied how defoliation stress affected the N contents of major plant organs in late autumn, N losses during the winter and N accumulation in the following spring. Plants were established from stolon cuttings and transplanted to pots that were dug into the field at Apelsvoll Research Centre (60 degrees 42'N, 10 degrees 51' E) and at Holt Research Centre (69 degrees 40' N, 18 degrees 56' E) in spring 2001 and 2002. During the first growing season, the plants were totally stripped of leaves down to the stolon basis, cut at 4 cm height or left undisturbed. The plants were sampled destructively in late autumn, early spring the second year and after 6 weeks of new spring growth. The plant material was sorted into leaves, stolons and roots. Defoliation regime did not influence the total amount of leaf N harvested during and at the end of the first growing season. However, for intensively defoliated plants, the repeated leaf removal and subsequent regrowth occurred at the expense of stolon and root development and resulted in a 61-85% reduction in the total plant N present in late autumn and a 21-59% reduction in total accumulation of plant N (plant N present in autumn + previously harvested leaf N). During the winter, the net N loss from leaf tissue (N not recovered in living nor dead leaves in the spring) ranged from 57% to 74% of the N present in living leaves in the autumn, while N stored in stolons and roots was much better conserved. However, the winter loss of stolon N from severely defoliated plants (19%) was significantly larger than from leniently defoliated (12%) and non-defoliated plants (6%). Moreover, the fraction of stolon N determined as dead in the spring was 63% for severely defoliated as compared to 14% for non-defoliated plants. Accumulation in absolute terms of new leaf N during the spring was highly correlated to total plant N in early spring (R-2 = 0.86), but the growth rates relative to plant N present in early spring were not and, consequently, were similar for all treatments. The amount of inorganic N in the soil after snowmelt and the N uptake in plant root simulator probes (PRS (TM)) during the spring were small, suggesting that microbial immobilisation, leaching and gas emissions may have been important pathways for N lost from plant tissue.

A new compact wastewater treatment system for use in single houses has been constructed in eastern Norway. The system is based on the principles of sub-surface flow constructed wetlands using various types of Filtralite as filter media. It consists of a septic tank followed by an aerobic biofilter succeeded by an upflow saturated filter. The aerobic biofilter is essential to remove organic matter and achieve nitrification, while the upflow filter polishes the wastewater and removes microorganisms and phosphorus. During the first 3 years of operation, the system has show stable and high removal with the following average values measured from the outlet of septic tank to the outlet of the upflow filter: 97.0%-BOD7, 30%-N, 99.4%-P, and 70.8%-SS. No Escherichia coli or somatic coliphages have been detected in the effluent. Due to considerable removal of organic mater, nutrients, and pathogens, the effluent will not negatively affect water and soil ecosystems. The system requires low maintenance and is designed to remove phosphorus for 5 years before renewal of the upflow filter media. When saturated with phosphorus, the media is a suitable fertilizer for plant production. (c) 2006 Elsevier B.V. All rights reserved.

In this study, forest treated with different strengths of selective cutting (2570 % of volume removed) was remeasured after twelve years in 2005. At each of the sixteen 0.2 ha plots, including four repeats of each treatment, all trees larger than dbh 2.5 cm were calipered. We investigated growth, changes in stand structure, tree age, tree damage and crown condition.The diameter distribution displayed a reverse J-curve at all plots both before and after the cuttings. Twelve years later the curve is only slightly changed. Increment cores from 300 trees were taken to analyse annual growth reactions in different diameter classes. Most trees reacted with increased growth from the second or third year after the cutting.This improved growth accelerated the following six or seven years with 20-80% increase. Both small and large trees reacted, including severely suppressed trees. The initial crown volume and crown vitality after cutting is essential for the increased growth since several years are necessary to build up a larger and better crown. The relationship between increased growth and reduced volume per hectare indicates less competition between trees regarding nutrients and light after the cutting.

Transportation planning is an important part of the wood flow chain in forestry. There are often several forest companies operating in the same region and co-ordination between two or more companies is rare. However, there is an increasing interest in collaborative planning as the potential savings are large, often in the range 5-15%. A key question is how savings should be distributed among the participants. In this paper we investigate a number of possibilities based on economic models including Shapley value, the nucleolus, separable and non-separable costs, shadow prices and volume weights. We also propose a new allocation method based on finding as equal relative profits as possible among the participants. A case study including eight forest companies is described and analyzed.

The storm Gudrun hit southern Sweden in January 2005 and approximately 70 million cubic meters of forest was wind felled. The existing logistic planning at forest companies in the damaged area had to be changed over night. There was a direct shortage of both harvest and transportation capacities. Key questions that arised were which terminals to use, where to harvest, where to store, which transportation modes (truck, train, ship) to use. In this paper we describe how the forest company Sveaskog made use of Operations Research (OR) as an important decision support in their supply chain planning in the aftermath of the storm.
